Single-pool Kt/V (spKt/V) assumes urea is distributed in a single, well-mixed compartment. The Daugirdas formula (2nd generation) is the most widely used: spKt/V = -ln(R - 0.008 × t) + (4 - 3.5 × R) × UF/W, where R = post-BUN/pre-BUN, t = session time in hours, UF = ultrafiltration volume in liters, and W = post-dialysis weight in kg. Minimum target is 1.2 per session. The simplified calculation makes spKt/V practical for routine monthly monitoring. However, it overestimates true dialysis dose because it ignores urea rebound — the post-dialysis rise in BUN as urea redistributes from poorly dialyzed compartments (muscle, bone) into the bloodstream.
